I used one of the ProV1 test balls for 53 holes(lost it on the 54th) and it seemed to be like a new ball each strike entirely. I use only the ProV1's because having the ability to try and test a sleeve of several brand name balls at the same time and hole, I can give them each the same external factors equally. My question is, I have noticed that during some rounds it seems that later in the round the ProV seems to feel softer off the driver. Now could this be the repeated club hits on the ball or could it be loosing the elasticity or core firmness? Soft spots?

I don't think it has anything to do with the elasticity of the core or the formulation of the urethane cover. It may be that you are loosened up and running on slight adrenaline or possible swinging smoother and making consistent contact with the sweet spot of your clubs. I play the 2015 Pro V1 along with a full compliment of Titleist clubs do not notice these inconsistencies that you mention.
